Targeting N-heterocyclic carbenes (NHCs) with increased π-acceptor character featuring N-fluorophenyl substituents, the molecular 2-chloro-1,3-bis(fluorophenyl)imidazolidine-4,5-diones (1a–c) were isolated from the condensation of the corresponding formamidine with oxalyl chloride. A hydroxypropylcellulose–succinic anhydride (HPC–SAn) conjugate was synthesized homogeneously at 80 °C for 6 h under N2 in N,N-dimethylacetamide (DMA). The HPC–SAn conjugate was further covalently linked with salicylic acid (SA) drug using the versatile reagent ZrOCl2·8H2O at 80 °C for 6 h. Multiple crosslinking of the benign HPC–SAn–SA conjugate was achieved using oxalyl chloride. The resultant cross-linked prodrug (CL-HPC–SAn–SA conjugate) was characterized using different spectroscopic techniques. In this work, the most discriminating synthesis-related impurities found in samples from seizures and controlled synthesis of the synthetic cannabinoid MDMB-CHMICA (methyl (S)-2-(1-(cyclohexylmethyl)-1H-indole-3-carboxamido)-3,3-dimethylbutanoate) were characterized. Based on 61 available powder samples of MDMB-CHMICA, 15 key-impurities were assessed, isolated in larger quantities via flash chromatography and structurally elucidated and characterized via high resolution mass spectrometry and nuclear magnetic resonance spectroscopy. Synthesis and biological evaluation of structural variants of carbazoquinocin C

Some new structural variants of the alkaloid carbazoquinocin C were synthesized in a few steps with good to excellent yields. The key step comprises a cyclisation reaction of appropriate 2-vinylindoles with oxalyl chloride. The carbazole-3,4-quinones are able to trap oxygen-centred radicals. In some biological/biochemical assays some of these compounds exhibit extraordinary results including inhibition of cyclooxygenase-1 and 5-lipoxygenase in the μM-range. Moreover some of the carbazoquinocin C-variants inhibit significant oxidative damage of cellular DNA in nM-range.

2-(Triphenylphosphonio)ethoxycarbonyl-(Peoc-)aminosauren 1 und -hydroxysauren 5 werden mit Oxalylchlorid in bei Raumtemp. stabile Saurechloride 2 bzw. 6 ubergefuhrt. Mit diesen aktivierten Verbindungen werden sterisch gehinderte Amidbindungen — z. B. in den geschutzten Valin-Peptiden 3a—g und in Hydroxyacyl-Dipeptiden 7 — hergestellt. Nach Schutzgruppenabspaltung aus der voll geschutzten Form 7a entsteht so die Sporidesmolsaure B (9a), die in ihren Eigenschaften mit dem Abbauprodukt aus dem Sporidesmolid I ubereinstimmt.
